Once I finish an approved online 5-hour course, how quickly does the DMV system update my record? I want to know if I can book my road test immediately or if I should wait a few days for processing.

In most cases the update doesn't take very long, but it depends on how the course provider reports your completion to the DMV. I would still complete the course as early as possible so you have enough time to schedule the first available road test without unnecessary delays.
